Position Description


The Research Scientist will participate in research projects that focus on the generation and characterization of novel Chimeric Antigen Receptor T cells (CAR-T cells) for use in the treatment of children with high-risk leukemia and solid tumors. The projects include the development of off-the-shelf, universal CAR-T cells, phenotypic characterization of CAR-T cells expanded under different conditions, and molecular profiling of the tumor-immune microenvironment surrounding pediatric solid tumors.


The Research Scientist will process immune cells collected from humans, transfer immune-modulating genes into the cells, and evaluate the tumor-killing capacity of the cells in model systems. The Research Scientist will review the goals and methodologies of the research projects with the Principal Investigators (PIs), design and implement research plans, perform experiments, and compile and analyze data for manuscript publication and grant submission. The Research Scientist is expected to be able to work both independently and collaboratively with other researchers and laboratory personnel.


The Research Scientist will be responsible for managing the daily activities of the laboratory, including general facility maintenance, ordering of supplies, control of inventory, and upkeeping of equipment. The Research Scientist will be responsible for training and supervision of laboratory technicians and trainees, with assistance from the PIs and other senior scientists. The Research Scientist will be responsible for the development and validation of research protocols, including, but not limited to, immunological techniques like antibody staining, flow cytometry analysis, cytotoxicity assays, expansion and characterization of immune cells, multiplex immunofluorescence imaging studies, and molecular biology techniques like DNA cloning, cellular transfection, lentivirus-mediated gene transfer, and single-cell RNA sequencing.


The Research Scientist will be responsible for the maintenance of a small colony of mice for use in the development of xenograft mouse models of leukemia and solid tumors and will perform procedures such as intravenous injection of human immune cells, subcutaneous implantation of tumor cells, and in vivo bioluminescence imaging of mice. The Research Scientist will work closely with core research facilities such as the Good Manufacturing Practice (GMP) Laboratory, Blood & Marrow Transplant Laboratory, Flow Cytometry Laboratory, and Genomic Sequencing and Bioinformatics Cores. The Research Scientist will assist the PIs in preparing data for manuscript publication, grant submission, IRB/IBC/IACUC approval, and FDA application.


The Research Scientist will be familiar with the GMP laboratory processes that are required and capable of drafting and following Standard Operating Procedures for the preparation of clinical-grade cellular products for testing and experimentation.
